ABSTRACT

Introduction: Aluminium, a ubiquitous metal implicated in some neurodegenerative diseases is linked to activation of free oxygen species. The antioxidant-rich plants, Moringa oleifera (MO) is reported to protect against Aluminium activities. This study investigated the actions of MO leaf extract (MOLE) against Aluminium chloride (AlCl3)- induced hippocampal cellular changes and serum levels of alkaline phosphatase (ALP), aspartate transaminase (AST) and alanine transaminase (ALT) in adult Wistar rats.Materials and Methods: Thirty Wistar rats weighing between 150 g and 220 g were grouped (n=5) into; 1-control (5 mL/kg distilled water), 2-AlCl3 (100 mg/kg), 3-low dose MOLE (250 mg/kg), 4-high dose MOLE (1,000 mg/kg), 5-concurrent AlCl3 and low dose MOLE, and 6-concurrent AlCl3 and high dose MOLE. All administrations were by oral gavages for 21 days. On day 22, following deep anaesthesia and cardiac puncture, blood was obtained for serum enzyme analysis, and the brain perfusion fixed, harvested and processed for histological study.Results: Results showed significantly (p < 0.05) higher ALP level in the AlCl3 group compared with the control, as well as the other test groups. However, there was no significant (p > 0.05) AST and ALT levels. The hippocampal CA3 of the AlCl3 group showed hypertrophic cells, with some of the cells having karyorrhectic features. The concurrent AlCl3 and low and high doses, MOLE groups showed less of these adverse features.Conclusion: These results suggest that MOLE may protect enzymatic activities against Aluminium chloride. However, its action on hippocampus is still subject to further investigation.


Introducción: El aluminio, un metal presente en diversos lugares implicado en algunas enfermedades neurodegenerativas, está relacionado con la activación de especies reactivas de oxígeno. Se informa que las plantas ricas en antioxidantes, Moringa oleifera (MO) protegen contra la acción del aluminio. Este estudio investigó las acciones del extracto de hoja de MO (MOLE) en los cambios celulares del hipocampo inducidos por el cloruro de aluminio (AlCl3) y los niveles séricos de fosfatasa alcalina (ALP), aspartato transaminasa (AST) y alanina transaminasa (ALT) en ratas Wistar adultas.Materiales y métodos: SE utilizaron treinta ratas Wistar divididas en 5 grupos, los animales pesaban entre 150 gy 220 g; 1 control (5 ml / kg de agua destilada), 2-AlCl3 (100 mg / kg), 3 MOLE de dosis baja (250 mg / kg), 4 MOLE de dosis alta (1000 mg / kg), 5 AlCl3 concurrente y MOLE de dosis baja, y MOLE 6-concurrente y MOLE de dosis alta. Todas las administraciones fueron por sonda oral durante 21 días. El día 22, después de la anestesia profunda y la punción cardíaca, se obtuvo sangre para el análisis de las enzimas séricas y la perfusión cerebral se fijó, recogió y procesó para el estudio histológico.Resultados: Los resultados mostraron un nivel de ALP significativamente (p <0.05) más alto en el grupo AlCl3 en comparación con el control, así como en los otros grupos de prueba. Sin embargo, no hubo niveles significativos (p> 0.05) de AST y ALT. El hipocampo CA3 del grupo AlCl3 mostró células hipertróficas, y algunas de las células tenían características cariorrecticas. Los grupos de AlCl3 concurrentes y dosis bajas y altas, MOLE mostraron menos de estas características adversas.Conclusión: Estos resultados sugieren que MOLE puede proteger las actividades enzimáticas contra el cloruro de aluminio. Sin embargo, su acción sobre el hipocampo aún está sujeta a más investigaciones.

ABSTRACT

ObjectiveTo explore the effects of hemofiltration on serum enzyme (SE),endotoxin (ET) and malonaldehyde (MDA) of dogs with heat stroke caused shock.MethodsSixteen healthy male hybrid dogs were randomly divided into 2 groups,8 for each:as heat stroke group (HS group ) and hemofiltration group (HF group).Severe heat stroke model was induced with high temperature.The dogs were taken out of the heating cabin when it reached heat stroke level,and then observed under normal temperature without treatment.The dogs in HF group was immediately treated with hemofiltration.The changes of SE,ET,MDA of two groups of dogs were observed and the survival time between two groups was compared,ResultsThe time from heat exposure to shock was ( 107.00 ± 28.52 ) and ( 111.38 ± 22.24 )minutes in HS group and HF group respectively ( t =- 0.354,P =0.729 ).The SE ( CK,LDH,ALT,AST) of the dogs were all higher after heat stroke,and the dogs of two groups showed no siginificant difference (P > 0.05).At three hours after heat stroke,the SE increased apparently in HS group and HF group,but the level was significantly lower in HF group. Before heat stroke,the serum ET showed no siginificant difference between two groups ( P > 0.05 ).After heat stroke,the serm ET was much higher than before ( P <0.01 ),but there was still no siginificant difference between two groups ( P >0.05 ).At three hours after heat stroke,the ET increased both in HF group and HS group,but the level was lower in HF group.Before heat stroke,the serm MDA had no siginificant difference between two groups ( P > 0.05 ).After heat stroke,the serm MDA was much higher than before ( P < 0.0l ),but there was still no siginificant difference between two groups (P > 0.05 ).After heat stroke in three hours,the MDA of HS group rose apparently while HF group slowly declined.The median survival time of HF group was 180 min while HS group was 75 min,the survival rate showed siginificant difference (P < 0.01 ).Conc4usions HF can improve the prognosis of dogs with heat stroke caused shock,prolong its survival time,reduce mortality.The mechanism is probably that HF clear serum MDA,partially clear serum ET and then eventually reduce cell and tissue injury and reduce SE.

ABSTRACT

Objective To observe the therapeutic effects of the co-administration of γ-aminobutyric acid (CABA), sodium dimercaptopropane sulfonate (Na-DMPS) and vitamin B6 in large doses on liver and heart of rats with acute tetramine intoxication, and compare their separate effects of either GABA or Na-DMPS alone with those of the triad combination. Method Thirty rats were randomized into control group (n = 6), tetramine intoxication without treatment group (n = 6), tetramine intoxication treated with GABA group (n = 6), tetramine intoxication treated with Na-DMPS group (n = 6) and tetramine intoxication treated with triad combination (GABA + Na-DMPS + vitamin B6, GNDV n = 6) group. Samples of blood, liver tissue and heart tissue of rats with acute tetramine intoxication were collected immediately two hours after medication with different drugs. Serum alanine aminotrasferase (ALT), aspartate aminotransferase (AST), creatine kinase (CK) and creatine kinase isoenzyme (CK-MB) were measured, and the pathological changes of liver tissue and heart tissue were observed under microscope. Results The symptoms of poisoning were apparently relieved and the latency for convulsion/muscular twitch were obviously delayed in poisoned rats treated with GABA, Na-DMPS and GNDV separately. Furthermore, combination group showed the latent period delayed longer than either GABA or Na-DMPS groups The GABA, Na-DMPS and GNDV significantly lowered the serum levels of ALT, AST, CK and CK-MB in rats with tetramine intoxication, and those serum levels of enzymes were lower in GNDV group than those in either GABA group or Na-DMPS group. However, there were no difference in those serum enzymes between GABA group and Na-DMPS group. Moreover, the intoxicated rats treated with combination treatment had the slightest pathological changes in liver and heart (GNDV < GABA or Na-DMPS). Conclusions The co-administration of γ-aminobutyric acid, sodium demercaptopropane sulfonate and vitamin B6 in large doses for the treatment of tetramine intoxication is a method of choice.

ABSTRACT

The changes of the activities of serum CK,LDH,HBKH and AST of 11 soldiers having stayed in simulated 3500m and 4500m were observed.It was found that the activities were significantly increased in the order of CK,LDH and HBDH because of hypoxia but the changes of AST was of no statistical significance.CK/LDH ratio and pulse rate showed similar charges to serum enzymes but the changes of arterial oxygen saturation and working efficiency were the opposite to those of serum enzymes.These findings suggest that serum CK and LDH are the indicators of choice to evaluate human reactions to high altitude.
